A new PCR approach for the identification of Fusarium graminearum BJM

The main objective of this work was to develop a PCR protocol for the identification of Fusarium graminearum, based on a pair of primers targeted to a segment of the 3' coding region of the gaoA gene that codes for the enzyme galactose oxidase (GO). This region has low homology with the same region of GO genes from other fungi. Genomic DNA from 17 strains of Fusarium spp. isolated from diseased cereals, from several other Fusarium species, and from other fungi genera was analyzed in a PCR assay using this primer set. Ochratoxin A (OTA) is one of the most abundant food-contaminating mycotoxins. Its occurrence in several agricultural commodities has been considered a worldwide problem. This toxin is mainly produced by Aspergillus species. OTA has nephrotoxic, immunosuppressive, and carcinogenic effects and consequently the contamination with this toxin represents a high risk for human and animal health. Characterization of Some Efficient Cellulase Producing Bacteria Isolated from Pulp and Paper Mill Effluent Contaminated Soil BABT

ABSTRACT The wide variety of bacteria in the environment permits screening for more efficient cellulases to help overcome current challenges in biofuels production. This study focuses on the isolation of efficient cellulase producing bacteria found in pulp and paper mill effluent contaminated soil which can be considered for use in large scale biorefining. Four different bacterial strains were isolated and screened for cellulase production by using CMC agar medium. All isolates showed cellulase activity these strains were further characterized by morphological, physiological, biochemical and 16S rRNA gene analyses. These isolates were identified as two Bacillus subtilis sub sps, Bacillus mojavensis and Bacillus cereus.

ABSTRACT Bovine digital dermatitis (BDD) is an infectious and contagious disease characterized by ulcerative and proliferative lesions affecting the skin on the bulbs of the heel or the interdigital cleft in dairy cattle, often associated with lameness. Evidences on the etiology of BDD indicate that it is multifactorial, involving environmental factors and multiple bacterial colonization. ABSTRACT The purpose of this study was to identify the yeasts involved in spontaneous fermentation of cocoa from the Brazilian Amazon region. The fermentation process was carried out experimentally with cocoa seeds from two sites (Medicilândia and Tucumã), State of Pará, northern Brazil, during a six-day period. Efficient detection of Frankliniella schultzei (Thysanoptera, Thripidae) by cytochrome oxidase I gene (mtCOI) direct sequencing and real-time PCR BABT

ABSTRACT Identification of Thysanoptera is based mainly on external morphology examination that can be time-consuming and difficult for non taxonomic experts. In this work, we propose a rapid and efficient molecular method to identify Frankliniella schultzei, an important and widespread pest thrips vector of tospoviruses in South America countries. Species-specific primers designed in the mitochondrial cytochrome oxidase I gene (mtCOI) were optimized for detection by conventional PCR and real-time PCR. The primers were tested on immature and adult thrips collected from crops and weeds found in São Paulo State. All samples collected were identified as F. schultzei, indicating the high prevalence of this species as vector of tospoviruses in Brazilian fields.

Evaluation of the genetic diversity among maize lines (Zea mays L.) by DNA fingerprinting analysis Ciência Rural

Informativo DNA fingerprint profiles of eight homozygotic maize lines were obtained by the electrophoretic separation of DNA restriction fragments and the ir hybridization with the minisatellite probe R18.1. The analysis of the bandsharing frequencies allowed to identify all the lines and to estimate the genetic distances between them. The relationship obtained by DNA fingerprinting analysis of the eight inbreed lines was highly consistem with the ir genetical origin.

Fungal endophytes of Panicum maximum and Pennisetum purpureum: isolation, identification, and determination of antifungal potential R. Bras. Zootec.

ABSTRACT This study aimed to isolate and identify endophytic fungi from the forage grass P. maximum and evaluate their ability to inhibit the growth of plant pathogenic fungi. One sample from P. purpureum grass was also included. Surface disinfected stem fragments were used for endophytic fungal isolation. One hundred and twenty-six endophytic fungi were isolated, of which 118 were from P. maximum and eight from P. purpureum. Morphological characteristics and internal transcribed spacer (ITS) and 18S (NS) sequence comparisons identified most isolated endophytic fungi as belonging to the phylum Ascomycota, with Sarocladium being the dominant genus. El género Colletotrichum es considerado uno de los principales hongos patógenos de plantas, este hongo se encuentra distribuido a escala mundial y afecta a diferentes cultivos tanto en pre y postcosecha. En el presente trabajo se aislaron e identificaron morfológicamente y molecularmente ocho aislamientos de Colletotrichum sp, de banano, papaya, carambolo, chile manzano, aguacate, mango ataulfo y mango manila, estos se inocularon sobre cada uno de los frutos para evaluar el fenómeno de infección cruzada. The objective of this study was to verify the prevalence of Babesia species (spp.) using molecular methods in dogs living in urban and rural areas of Cuiabá, Mato Grosso State, Brazil, and to identify the main factors associated with infection. ABSTRACT: Soybean is a commodity of great economic importance worldwide, particularly in Brazil, world’s second largest producer. Nematodes, especially those of the Meloidogyne genus, severely limit productivity. Identification of nematode species is important for effective soybean management. Here, 26 populations of root-knot nematode (Meloidogyne spp.) from 15 municipalities in the states of Bahia, Mato Grosso, Goias, and Minas Gerais were characterized based on the morphology of the female perineal region, esterase profile, and identification based on amplification of specific regions of the population genome. New isolates of Trichoderma antagonistic to Sclerotinia sclerotiorum Biota Neotropica

Abstract Forty-nine isolates of Trichoderma from the Brazilian Midwest were evaluated for their antagonistic activity in vitro against Sclerotinia sclerotiorum (causal agent of white mold), which were then identified based on their nuclear ribosomal ITS sequences. Paired culture tests showed that all isolates exhibited some antagonism, with a maximum of 77% mycelial inhibition and complete inhibition of sclerotia production. Two isolates were found to be the most promising biocontrol agents, considering both antagonistic parameters (CEN1253 - T. koningiopsis and CEN1265 - T. brevicompactum). Background: Corsica is a mountainous French island in the north-western Mediterranean presenting a large diversity of natural environments where many interactions between domestic animals and wild fauna occur. Despite a favourable context for ticks and tick-borne diseases (TBDs), the tick fauna of Corsica has not systematically been investigated. Methods: For one year (May 2014-May 2015), a survey of ticks infesting cattle was performed in the three Corsican cattle slaughterhouses. Two of these were visited monthly and one quarterly; the ticks were manually collected, just after flaying.
